Resumo

Obtaining bioactive substances from natural products is a concern of modern medicine. For this, biotechnology provides some tools such as calli in vitro culture, to obtain substances of interest. Current studies with Deguelia duckeana revealed anticancer potential of flavonoids isolated from the roots of the species. In this work, we initially aimed to establish and in vitro culture of the plant species. Once callus obtained, they were dried and extracted with hexane and ethyl acetate. The extracts were fractionated to isolate the substances produced by the callus. As results obtained are that: in vitro callus establishment of this species was possible, aseptic cultures were obtained from young leaves of D. duckeana that underwent a disinfestation process, followed by induction and multiplication in MS medium added of 3 mg.L-1 of BAP + 2 mg.L-1 of NAA + mg.L-1 of 2 Kin. The calli were dried in lyophilizer and the 1: 1 combination of the hexane/ethyl acetate solvents proved to be the best for extracting the present metabolites. Fractionation of hexane/ethyl acetate 1:1 extract from callus allowed to isolate the triterpenes 3β-hydroxy-20(29)-lupen-28-al (betulinaldehyde) and lupeol. Their structures were confirmed by analysis of 1H and 13C NMR spectra, and COSY, HSQC and HMBC contour maps.
