Resumo

Cellulases are hydrolases, with great interest in industrial, notable for its power in the production of ethanol by hydrolysis of cellulolytic materials. The actinobacteria are producing this enzyme, especially of the genus Streptomyces. This study aimed to produce and characterize a portion of a cellulolytic complex excreted by actinobacteria belonging to the microorganisms Collection of UFPEDA. In the selection of microorganisms producers of cellulolytic complex in qualitative testing microorganisms 171x and 178 showed the most significant enzymatic Index. As for the complex production time, fermentation was studied by 7 days with aliquots being taken every 24h. There was thus obtained peak production at 48 the fermentation was also noted that the micro-organism 171x enzyme produced in greater quantities. The experiment followed with only the more significant production actinobacteria (171x), was tested for optimum substrate concentration (carboximeticelulose), growing temperature and an initial pH of production. The results demonstrate greater production of the cellulolytic complex with a substrate concentration of 3%, fermentation temperature 45 ° C and initial pH 4.0. About the partial characterization of the complex maybe that this behaved very satisfactorily, as proved to be very stable during the test. Furthermore, it has proven activity of the complex by zymography method and its probable molecular weight, the cellulolytic complex studied have Michaeliano behavior, the enzymatic hydrolysis product suggests that the enzyme complex is composed of endoglucanases and experiment suggests that the complex should be produced at 40ºC, pH 4 and 2.5 g / L of CMC. The micro-organism was identified at genus level by the microculture and kind method, molecular identification, being identified as Streptomyces capoamus. Data from this study showed the importance of isolated actinobacteria biotechnology of the savanna soil. This complex has potential for use in the treatment and environmental cleaning products industries because it has high stability in adverse conditions
