Resumo
Cutaneous leishmaniasis is an infectious disease and can be caused by protozoan of the genus L. guyanensis, with different clinical manifestations. Infection occurs by inoculation of the promastigotic form metaclyclic into the host body during the blood supply of the infected female, triggering immune response. Interleucine 12 (IL-12) It is key cytokine for the development of TH1 cells, response TH1 is known for the elimination of intracellular microorganism, macrophage polarization (M1), IFN-γ increase, LC resistance. Case-controle study, to investigate variants, present through the IL12B gene in LC patients (case group) and healthy individuals (control group) without leishmaniasis history, from the same endemic areas of the case group. IL12B variants analyzes were performed by PCR-RFLP and cytokine dosage (IL-12 and IFN-γ) by the Luminex method. From the results obtained, it was seen that the presence of the SNV C/C genotype RS2569253 was associated with LC susceptibility (p = 0.32; or = 1.53 [95% ic 1.07-2.22], in contrast, genotype A/G da SNV RS3181224 was associated with LC protection (p = 0.032; or = 0.68 [95% IC 0.51-0.91]. The connection imbalance of SNV rs3181225 with rs919766 (R2 = 0.85), and Haplotype H3 C/A/A/A/ is associated with susceptibility compared to LC (OR 1.6; 95% [1.20-2.27]; p = 0.001). It is also seen that the genotype c/c, snv RS2569253, (C/C vs T/T) It was correlated with low production of IL-12 (p = 0.05) in the control group. Thus, these results suggest that the variants present in the IL12B gene may have an important influence on the clinical outcome of LC.
