Resumo

The goal of this study was develop a protocol of in vitro propagation of andiroba, copaiba, breu branco and rosewood, trough apical, leaves and petiole obtained from seedling. The seeds were treated by immersion for 24 hours in derosal (5ml.L-1), 30 minutes in sodium hypochlorite (0.85%), and then germinated in trays filled with washed sand and vermiculite. To explants in vitro establishment were tested 4 asepsis protocols composed of derosal (5ml.L-1), NaOCl (0.85%), ampicilin (500mg.L-1), cloraphenicol (500mg.L-1) and tween 20 (1ml.L-1), with 50 repetitions per treatment. To induce in vitro organogenesis was used MS medium supplemented with NAA, IAA, BAP and TDZ totalizing 24 treatments and 1 witness, with 10 repetitions each, the Tukey test was applied in 5% of significance. After the inoculation in flood hood chamber the explants were kept in B.O.D. with temperature of 30oC for three days, then taken to growth room with 16 hours of photoperiod, 25μ moles.m-2.s-1 radiation and 30oC temperature. To acclimatizing were tested 6 substrates (treatments) composed of combinations of washed sand, organic compound and clay with 10 repetitions. All the explants of andiroba, breu branco and rosewood were established in rates bigger than 81% with application of a protocol composed of ampicilin (500mg.L-1) and derosal (5ml.L-1), plus NaOCl (0,85%) and tween 20 (1ml.L-1). There was regeneration of apical source of andiroba, breu branco and rosewood trough application of NAA and BAP. Leaves and petiole of andiroba, breu branco and rosewood rooted with NAA supplementation, but there weren’t shooting formation. Wasn’t possible establish explants of copaiba.
