Avaliação das condições de fermentação semissólida na produção de protease coagulantes por Aspergillus flavo-furcatis (Batista & Maia 1956)

Resumo

Thirteen strains of Aspergillus flavofurcatis were subjected to semi-solid fermentation of Amazonian agro-industrial waste, açaí seed and cupuaçu peel, supplemented with rice bran, at 25 ºC in 5 days. Proteolytic activity was performed using 1% (w/v) azocasein as substrate in buffers under standardized conditions. The enzyme activity was determined in 100% of the extracts. The A. flavo-furcatis strains expressed proteolytic activity in both agroforestry residues. In açaí seeds supplemented with rice bran, the average proteolytic activity was 46.32% higher than that determined in cupuaçu and rice bran (465.50 U/mL). Among the Aspergillus evaluated, a significant protease activity (929.2 U/mL) was expressed by A. flavo-furcatis DPUA 1608 when açaí seed was used in the bioprocess. In cupuaçu peel, Aspergillus flavo-furcatis DPUA 1539 was the one that expressed the highest activity (619.77 U/mL). The cultivation conditions that provided a significant increase in proteolytic activity were inoculum with 10 days of cultivation, fermentation in five days . A. flavo-furcatis strains are promising sources of proteases for future industrial application when cultivated in the Amazonian agroforestry residue studied.
