Resumo

Natural environment has suffered high accumulations of pollutants which not only affect the soil, groundwater, rivers, lakes, water courses and air, but also human health due to forming of unhealthy conditions. Among the many effects when crude oil and derivates spill, the released xenobiotics to environment make worrisome because of its enviromental persistence and harmful effects to life. One solution to this problem is bioremediation, a tool that uses microorganisms for biochemical degradation of these compounds, transforming them into less or non-toxic substances. In this context, a previous study isolated the bacteria A. junii SB132 from macrophytes of the Negro River bank located by the port of Manaus and confirmed its identity by morphology and 16S gene sequencing. Biodegradation tests performed in laboratory have confirmed the ability of this bacteria to degrade oil and diesel fuel. Therefore, the aim of this research was to identify proteins of A. junii SB132 related in degradation pathways of the crude oil and diesel fuel. Ribosomal proteins, phenotypic, bacterial morphology, sensitivity profile to antibiotics and plasmid profile analysis were obtained. Proteins extracted from bacterial grown in crude oil, diesel fuel and yeast extract, 1.3% respectively, were used for 2-DE gels and similar and differential proteins were identified by using mass spectrometry and database at NCBI. The identity bacterial was confirmed and the plasmid profile was taken. These data revealed that the bacteria studied in this research is the really A. junii specie and that this had plasmids with size ranging 94.000pb to 5.000pb. Identified putative proteins were related to cellular processes, such as degradation of xenobiotic compounds, biosynthesis of molecules, DNA repair and protection, membrane transport proteins, cell signaling and transcription factors.
