Resumo

Penicillium are anamorphics fungi economically important for producing composts for uses such as supplement or food products improvement and also are used in bioremediation for environments recovery. Therefore, this work had as objective to verify the purity, authenticity, viability and the antimicrobial activity of 60 cultures of Penicillium from the Amazon region preserved in distilled sterilized water and under mineral oil, deposited in DPUA Culture Collection. The revised microorganisms were reactivated in Czapek Yeast Extract Agar (CYA) and authenticated based on macro and micromorphological characteristics on Malt Extract Agar (MEA), Glicerol Nitrate Agar 25% (w/v) [G25N] and Czapek Yeast Extract Agar (CYA). After authentication, the cultures were submitted to antimicrobial activity by the Gelose Block Method. The antimicrobial activity was performed in selective culture medium at 25oC and 37oC against the following test microorganisms: Candida albicans DPUA 1340, Staphylococcus aureus CCT 1352, Escherichia coli CCT 0547 and Mycobacterium smegmatis PDUFPE-7. The antimicrobial activity was evaluated by measuring the halo of inhibition against the test microorganisms. For biocomposts detection by bioautography plugs of cultures of Penicillium selected by diffusion in agar were submitted to the extraction in Ethyl Acetate (6: 4, v/v) using as standard Itraconazol and Riphampicine. Among the examined cultures, 90.0% expressed viability, purity and taxonomically confirmed in accordance with specialized literature. From these, 46.66% demonstrated positive result in the antimicrobial activity for diffusion in agar and 25.0% in the bioautographic assay. From the results it has proved preservation methods efficiency in distilled sterilized water and under mineral oil, as well as the Penicillium species potential in the biocomposts production with antimicrobial activity.
