Resumo

Staphylococcus aureus is the human pathogen more related to a disease acquired in the community and the hospital environment. This organism is extremely adaptable and over a short period of time has developed antimicrobial resistance strategies, thus, the bioprospection of new drugs capable of inhibiting methicillin resistant Staphylococcus aureus - MRSA is an important field for research development. In this recent context, in the laboratory of mycology - INPA has been identified a fungal strain,Paecilomyces H59 the production of substances capable of inhibiting MRSA. From this finding, the aim of this study was to investigate the production and characterization of an inhibitory component against MRSA produced by Paecilomyces H59. The methodological strategy to achieve this goal included:a) Taxonomic characterization of Paecilomyces H59;b) Optimization of the production of active substances by submerged bioprocess andc) Isolation and identification of substances active against MRSA.The taxonomic characterization of Paecilomyces H59 demonstrated all the fungal structures reported in the literature identifying it as belonging to the previously cited genus. The adjusted factors for the higher antimicrobial production were:a) Use of glucose as carbon source;b) Any source of nitrogen tested;c) pH 5; d) Inoculum 1x105 and absence of agitation.In the isolation and identification of active substances,a Eletronspray ionization mass spectrometry (ESI-EM) analyzes was performed in the two phases DCM and AcOEt (fractions 5-9 obtained with the dichloromethane / ethyl acetate system (20:80 v / v ) was performed comparisons of the spectra and found in both majority and similar peaks fractions with the mass of 155 m/z molecular formula C7H7O4, it is suggested that this is responsible for the antimicrobial activity against methicillin-resistant Staphylococcus aureus - MRSA. And check the in vitro cytotoxicity of the substance isolated by the Alamar Blue method. Results from this work indicate that the bioprospection of substances with antimicrobial activity produced by Paecilomyces H59 is an alternative for the research and development of new drugs with antimicrobial activity and is vital to protect human health against multiresistant bacteria.Patients, physicians and the pharmaceutical industry interest in this approach.
