Resumo

Shigella is the Gram-negative pathogen that causes acute dysentery, affecting mainly children under five years of age in low- and middle-income countries, causing thousands of deaths per year. As a result, several scholars have been working to search for an effective and safe alternative, mainly due to the lack of an effective vaccine and the spread of multiresistant strains. The peptide P9 (FimH) from Shigella flexneri was used in this study to immunize Gallus galus Dekalb White galiform to produce and evaluate IgY (Immunoglobulin Y). Anti-P9 IgY obtained and isolated by polyethyleneglycol precipitation methods and affinity chromatography was evaluated for its reactivity by the immunological techniques Dot blot, Western blot and Flow cytometry. IgY antiP9 was able to recognize the target antigen in the synthetic, denatured and native form of Shigella flexneri 5a M90T. The antibody obtained showed cross-reactivity against other strains of diarrheogenic bacteria, which can not be used for immunodiagnostic methods. On the other hand, these antibodies have great potential for use in passive immunity methods against Shigella spp.,And possibly serve as a basis for application enhancement against other diseases.
