Resumo
The amazonian species Arrabidaea chica Verlot (Bignoniaceae), known as crajiru, are useful in traditional medicine, because of antioxidant and anti-inflammatory potential. The aim of this study was to investigate the anti-aging and anti-inflammatory potential of five extracts from Arrabidaea chica species: crude aqueous extracts by infusion and decoction, ethylic acetate, hexanic and ethanolic. It was performed the cytokines quantification to evaluate the effect on inflammation, and was assessed the action against aging by intracellular reactive oxygen species (iROS) production, mitochondrial membrane potential, comet test and cytometry. The aqueous extracts reduced TNF-α (IC 50% = 35 μg/mL) and increased IL-10 (EC 50% = 31 μg/mL) with dose-dependent Zymosan in J774 macrophages, demonstrating the anti-inflammatory activity. The anti-aging ability was demonstrated in all the extracts by reduction of iROS production (p<0.0001) and enhancement of the mitochondrial membrane potential (p<0.0001) when compared to the staurosporine control in THP-1 macrophages. Furthermore, the extracts from Arrabidaea chica protected the double-stranded of DNA against doxorubicin action (p<0.0001) in THP-1 cells and protected the cells from apoptosis (p<0.0001), highlighting the major effect promoted by ethylic acetate extract (95% CI 83,60 – 123,1; p<0.0001) and aqueous extracts (95% CI 6,278 – 7,655; p<0.0001), respectively. The species Arrabidaea chica (crajiru) is easily accessible and we suggest that it should be explored as a therapeutic agent against cellular aging and inflammatory processes.
