Resumo
Duroia macrophylla is a Rubiaceae that has been highlighted with discoveries of the presence of secondary metabolites like alkaloids, flavonoids and terpenes. These metabolites have their importance in the exploration of molecules with therapeutic targets for the research of new medicines. Thus, the objective of this work is to analyze callus, leaves and branches of D. macrophylla Huber, aiming the knowledge of the proteins related to the production of the secondary metabolites through the proteomic analysis. In this work, the uni and bidimensional electrophoresis technique was used followed by identification of bands and spots by mass spectrometry. Of the 3,024 proteins identified in this study, 364 are of defense and response to stress and 51 of secondary metabolism in the three tissues studied. Among the proteins involved in the production of alkaloids, caffeine synthase and deacetoxyvindoline hydroxylase have been identified in callus, which is a catalytic enzyme for the production of vincristine and vincristine, belonging to the biosynthetic pathways of caffeine and indole alkaloids, respectively. This confirms that callus has the ability to produce secondary metabolites of pharmaceutical interest. This work described for the first time the proteomic analysis for callus, leaves and branches of D. macrophylla by means of mass spectrometry.
