Resumo

Endophytic fungi are useful resources for prospecting for active biomolecules. In this research, 4 fungi (BDL-DM5-G13; BDL-DM2-G15; SBL-DM1-G12; BDLDM8-G2) isolated from leaves of Duroia macrophylla, a plant species with marked biotechnological potential, were studied. Thus, the fungi were grown in potato dextrose broth with 0.2% yeast extract. The extracts and AcOEt phases of the mycelia were analyzed by comparative thin layer chromatography and hydrogen nuclear magnetic resonance, the presence of secondary metabolites of interest was verified, then the extraction fractionation was performed by open column chromatography. An evaluation of antioxidant activity was also carried out, as well as tests for toxicity against Artemia salina, antibacterial and antifungal activity. From the fractionation of the fungus extract (BDL-DM5-G13) identified as Graphium jumulu, the substance 4,9-dihydroxy 1,2,11,12-tetrahydroperylene3,10-quinone was isolated, which was identified through the analysis of the spectra uni and bidimensional 1H and 13C NMR, and mass spectrometry. For other fractionated extracts, it was not possible to perform the elucidation and/or structural identification of the substances limited by the mass and degradation of some samples. As for biological activities, only the extract (BDL-DM2-G15) was toxic to Artemia salina. For the diffusion antibacterial assay, all extracts showed average activity against seven tested bacteria. In the antifungal and antioxidant activity tests, none of the extracts were active. With the cultivation condition used, it was possible to verify that there was production of alkaloids, terpenes and quinones, being the species G. jumulu promising for chemical prospecting using other in vitro cultivation conditions, aiming at increasing the production of 4,9 dihydroxy-1,2,11,12-tetrahydroperylene-3,10-quinone, or other substances of interest.
