Resumo
Hepatitis C is a disease with a variable course where most cases result in chronic infection, which can lead to cirrhosis, hepatocarcinoma and liver transplantation. Only a minority of infected people are able to eliminate the virus during the acute phase without any therapeutic intervention. The factors explaining these differences are not yet well understood. In this sense, this study aimed to analyze the polymorphisms of IL-2 (rs2069762, rs4833248), and IL-2Rβ (rs1003694), and the relationship these polymorphisms with the production of cytokines of the profile Th1, Th2 and Th17 in patients infected with the Hepatitis C virus. In this study, 149 samples from individuals infected with the hepatitis C virus and 100 from healthy individuals were analysed. DNA extraction was performed using the Brazol® Kit. The determination of the allele frequencies of the polymorphisms was obtained by PCR-RFLP. The measurement of cytokines was performed by Flow Cytometry CBA (Cytometric Bead Array). Statistical analyzes were performed using the GraphPad Prism 5.0 program. The SNPs studied, were presented in Hardy-Weinberg equilibrium. No statistically significant were found in genotype and allele frequencies between the groups studied. The cytokines IL-2, IL-4, IL-6, IL-10, IL-17A, were increased in HCV + patients when compared to controls. The tumor necrosis factor was higher in the control group compared to the patient group. There was no statistically significant difference in the IFN-y level between the groups. Fibrosis> f2 was characterized by a high production of IL-2. The results obtained suggest the SNP IL2 rs4833248 and the SNP rs1003694 are not related to the development of Hepatitis C, the SNP IL-2 rs2069762 could be related to the chronicity of the disease and the high production of IL-2 in fibrosis > 2 is related to increased fibrogenic activity in patients with hepatitis C.
