Resumo
The drugs currently used come from various sources of natural products that are mostly extracted from animals, plants, bacteria and fungi. One of the ways to extract economic value is with the use of biotechnology by bioprospecting strategies for these resources. Well, it is known that the search for fibrinolytic enzymes with therapeutic applications to treat thrombosis has been one of the greatest demands of the pharmaceutical industry in recent times. Among the numerous organisms, mushrooms stand out for their ease of cultivation and high efficiency in the production of extracellular proteolytic enzymes. In this sense, this research aimed to carry out the production, pre-purification and biochemical characterization of proteolytic enzymes with fibrinolytic action of edible mushrooms. As a result of this dissertation, all specific objectives were met and are presented in three chapters, in the form of articles: Chapter I is characterized by a systematic review study, which concentrates an analysis of a survey carried out in the last ten years; Chapter II is characterized as an in vitro research where the Pleurotus eryngii species, an edible mushroom, was cultivated in two media by submerged fermentation in order to obtain an enzymatic extract; and chapter III, which is also characterized as an in vitro research where the enzymatic extract of the P. eryngii mushroom was pre-purified in two-phase aqueous systems. Based on these data, it is concluded that this in vitro study demonstrated that the enzyme of the edible mushroom Pleurotus eryngii has potential for use in pharmacological application as thrombolytic agents.
