Estudo químico de uma linhagem de Purpureocillium lilacinum com potenciais larvicida e antiplasmódico.

Resumo

The Purpureocillium lilacinum species is one of the most promising fungi strain at the source of bioactivity agent and is useful for biological control of nematodes. Has already been identified its action against several microorganisms, including bacterium, fungi, plants and insects. Thus, this project aimed the chemical and biological study of a Purpureocillium lilacinum strain, intending to extract and identify compounds that had antiplasmodial and/or larvicidal activity through bioguided study. The species was cultivated in PDY medium, and the extraction was carried out by partitioning the liquid medium with ethyl acetate and isopropanol in a 9:1 ratio, after 21 days of cultivation. The obtained extract was fractionated in C18 open-column chromatography and the resulting fractions were submitted to larvicidal assays against the Aedes albopictus vector and antiplasmodial assays against the protozoan Plasmodium falciparum, and the samples with positive results were analyzed by mass spectrometry and NMR. These fractions were then refracted in an open-column and HPLC, resulting in fractions rich in leucinostatins and another containing the configurational isomer of pinelic acid. Leucinostatin-containing samples showed high larvicidal and antiplasmodial activity, as well as high cytotoxicity against MRC5 fibroblast lineages
