Resumo
Cutaneous leishmaniasis is a non-contagious infectious disease, considered by the World Health Organization (WHO) as a neglected tropical disease (NTD). and even investments in scientific research and innovation for this disease affect society. Therapeutics involves the use of pentavalent antimonials (Sb5+), although several adverse effects are related. Furthermore, understanding the pathogenesis of leishmaniasis still represents a great challenge due to the diversity of vectors and species of the parasite involved in the infectious process. In this sense, the knowledge of the interaction with these agents and the understanding of the immunological mediators involved in the action are fundamental for the development of new therapeutic methods. Thus, this work aimed to evaluate the lymphoproliferation and production of cytoprokines from the supernatant of PBMC cultures stimulated by bioactives and soluble Leishmania stimulants (SLA) through in vitro assays. The lymph4 complex front against bioactive stimuli obtained from Libidibition Ferre, the LF, μgmL complex front, and the attribution to copper/something (281 μgmL), Glucantime® (100 μg/mL), L. amazonensis SLA (100 μg/mL), L. guyanensis SLA (100 μg/mL) and Phytohemagglutinin - PHA (10 μg/mL) using the BrdU Cell Proliferation ELISA Kit after 72h incubation. The quantification of IL-6, IL-8 and IL-1b cytokines was performed by BD™ Cytometric Bead Array (CBA) Human Th1/Th2/Th17 Cytokine Kit. The results can be used as a combination therapy. As for the antigens, both soluble Leishmania induced strong lymphoproliferation and increased levels of inflammatory cytokines, but there was only an increase in significance against the soluble Leishmania guyanensis.
